阿片类药物是临床上常用的镇痛药物。通过分子克隆已经鉴定出三种介导阿片效应的阿片受体(mu, delta和kappa)。药理学研究表明,每种类型的阿片受体由受体的亚型组成。虽然mu阿片类受体是介导阿片类镇痛作用的主要受体,但delta和kappa受体在抗痛觉中也很重要(例如,delta和kappa受体可以介导脊髓镇痛)。近年来,阿片类药物的细胞保护作用已被认识到。在缺血等有害事件中,阿片类药物的存在减少了包括心脏和大脑在内的多个器官的细胞损伤。在大多数研究中,这些影响似乎是由δ受体介导的。一种新的细胞保护形式,其中预先暴露于阿片类药物对细胞缺血(阿片类药物预处理)具有保护作用。在心脏中,这种阿片类预处理诱导的保护已被多项研究充分证明,可能由delta受体、G(i/o)蛋白、蛋白激酶C、atp敏感的钾通道和自由基介导。我们最初的研究表明阿片类物质预处理也能诱导神经保护。 This neuroprotection involves delta(1) receptors, mitochondrial ATP-sensitive potassium channels and free radical production. In this review, we will briefly describe the analgesic effects of opioids. We will focus our discussion on opioid preconditioning-induced protection and its mechanisms. Opioids and agents that specifically work on the signaling molecules for opioid preconditioning-induced protection may prove to be useful in inducing protection against ischemia in clinical practice.
